I am a few weeks away from getting my first chickens, i have a chicken coop inside a 10 ft x 5 ft steel galvanised fox proof run, do i still need to close the  pop hole at night ?

Could a fox dig underneath the run? If not, then as long as the door faces away from prevailing winds, then you should be ok. A few people down our allotment site don't lock their birds up at night and the birds are fine.

hopefully the foxes don't get in, i have buried mesh and laid 600 mm x 600 mm paving flags round the outside of the run, the fox would have to tunnel in, the pop hole is south facing, would rats be a problem?

It sounds like you have a similar set up to me, except I do like to lock the girls up at night no matter how late I get to them! Others have a rat problem, but they have old sheds adapted to chicken coops and use layers of chicken wire that rats can gnaw through. The steel mesh should prove difficult but not impossible for them to get through. Best thing is to clear food away each night as much as possible. My girls are messy and throw lots of pellets on to the floor of the run looking for the exact size they prefer  :) so this not always practical. Just keep your eyes open for signs of rats. Hopefully you won't be troubled by them, like me  :D

Can't  guarantee rats wont get in, if they did are they just after the food or will they harm the chickens?

You would have to be very unlucky for a cat to take an interest in adult chooks. However, mink as you say, and stoats and badgers cannot be dismissed. To leave the pop hole open you also need to consider if there may be a draught as this could be lethal to chooks. There is a difference between ventilation and a draught! :)
