Weedkillers such as glysophate are absorbed through the leaves, so you'll have to wait for regrowth if you follow that method.
Did it really tell you to do THAT on the packet?

Depends what weed killer, and what you intend to plant there and when.  Weeds need to be actively growing for things like glyphosate to work (e.g. round up) so if you strim first it will stop them working.  Other weed killers can stay in the soil for a long time so need to consider carefully before using.

When we had the same task we opted to dig, dig and dig some more to get the weeds out and all of the roots....has taken 2 years to get fully on top of it, but it was worth the back ache!

I've just done mine with roundup and they cleared well, i kept spaces covered whilst i did other areas then as i was getting nearer removed the cover and sprayed again. this gave me two week for it to die then i cleared that bit.
im two thirds of the way after two months but thats with building a shed and greenhouse etc so not so bad and ill be well ready to overwinter the beds which i have manured one for spud the rest ill plant green manure on.

Started off on a half-plot, and my method was dig, dig, dig.  Had a plot full of sky-high brambles.  So I chopped and chopped, then when it was clear I then dug out the roots.  I started in the Autumn, so I didn't bother growing anything that first winter.  I just dug out roots and smaller weeds, and then completely dug it over.  Was worth it in the end.  The bottom end of the plot was cleared, so I bought some gooseberry bushes (this was in the November/December), and they were my "hedge".  And it went from there and I've never looked back. :D
